  • Exploring the biomarker potential of THCAT155 gene with oncogenic function in papillary thyroid carcinoma

  • Introduction: Papillary thyroid cancer (PTC) is the most common type of thyroid cancer, and identifying the molecular factors involved in its development and progression is of great importance. Long non-coding RNAs (lncRNAs) play important roles in various cellular processes and cancer development through the regulation of gene expression. This project aimed to investigate the role and potential of THCAT155 as a biomarker in papillary thyroid cancer.
  • Methods: Transcriptomic data related to papillary thyroid cancer was obtained from The Cancer Genome Atlas (TCGA) database. Differential gene expression analysis was performed using the R open-source statistical computing environment, and genes exhibiting significant differences in expression between tumor tissues and adjacent normal tissues were investigated. Subsequently, the associations between gene expression levels and patients’ clinical characteristics and survival outcomes were evaluated. To further assess the potential of lncRNAs, selected genes were analyzed separately, and their expression patterns and reported roles across different cancer types were investigated. In addition, 20 paired PTC and adjacent normal tissue samples were collected from patients for future validation of THCAT155 expression using reverse transcription quantitative polymerase chain reaction (RT-qPCR).
  • Results: Among the selected genes, thyroid cancer-associated transcript 155 (THCAT155) was identified as the target of this study. Based on differential expression and survival analyses, this lncRNA showed a significant difference in expression between papillary thyroid cancer tissues and adjacent normal tissues (LogFC = 8.54, P = 8.66 × 10⁻⁷⁵). Given the limited available evidence regarding its function in thyroid cancer, THCAT155 may represent a promising candidate gene for further investigation of diagnostic biomarkers and the molecular mechanisms underlying PTC.
  • Conclusion: Considering the altered expression pattern of THCAT155 in papillary thyroid cancer, this lncRNA may have potential as an oncogenic biomarker in PTC.
