  • Association between rs6850524 Polymorphism in the CLOCK gene and susceptibility to breast cancer in Women from East Azerbaijan Province, Iran

  • Introduction: Breast cancer is a common malignancy in women , and circadian-clock genes have been investigated for their possible roles in cancer susceptibility . The CLOCK gene contributes to circadian regulation and has been studied in relation to breast cancer , although findings across variants and populations are not consistent . This study investigated the association between the rs6850524 polymorphism and breast cancer in a population from East Azerbaijan , Iran .
  • Methods: This case–control study included 200 participants : 100 women with breast cancer and 100 age-matched healthy controls . Genomic DNA was extracted from peripheral blood samples . DNA quality and concentration were assessed by spectrophotometry and agarose gel electrophoresis . The rs6850524 polymorphism was genotyped using polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) . The genotype and allele frequencies were compared between groups using the chi-square test in SPSS , version 23 . Statistical significance was set at P < 0.05 .
  • Results: The reported G-allele frequency was 59.5% in controls and 38.5% in patients ; the C-allele frequency was 40.5% and 61.5%, respectively . The supplied results additionally report a 21% higher G-allele frequency and a 21% lower C-allele frequency among patients , with P < 0.05 .
  • Conclusion: The findings as reported suggest a possible association between the CLOCK rs6850524 G allele and breast cancer in the studied population . However , the allele-frequency differences shown are small , and the stated 21% contrasts require verification .
